We have done a lot of work in the past month to reduce the size of users'
mailboxes. The sizes have been greatly reduced, yet I don't see much of a
change in available disk space. How do I reclaim that space. Is there a
defrag of the information store that I can do to recover it?

Also, I'm getting ready to migrate my exchange server to a new server. I
intend to do a disaster recovery method of migration using a backup tape.
Will everything come over as fragmented, or will it be almost like a fresh
install?

look in the application log for event id 1221 for source
MSExchangeISMailboxStore
this will tell you how much space is free. that is just an online defrag to
determine whitespace.
for example, if you have a 10gb store and this event says there is 1gb free
space, the actual used space is 9gb though the edb file is 10gb. to free up
the space you need to do an offline defrag. the store needs to be
dismounted and use the eseutil with the d switch for a defrag. (eseutil
under \program files\exchsrvr\bin) how long that takes will depend on your
hardware and size of the store.

Andy,

Before you run eseutil you should have a good backup of the Exchange server,
also make sure that you have free drivespace of at least 110 percent of the
size of the database being defragmented. You should read the following
Knowledge base articles to get a better understanding how eseutil works and
what the requirements are:
